Randomized patients with metastatic, nonsquamous, non-small cell lung cancer, previously untreated and lacking EGFR/ALK alterations, received either pembrolizumab (200 mg) or placebo, administered every three weeks for up to 35 cycles. This regimen incorporated pemetrexed and either carboplatin or cisplatin for four cycles, followed by continuous pemetrexed maintenance until disease progression or intolerable side effects were encountered. Progression-free survival (PFS) and overall survival (OS) were established as the primary evaluation criteria. A total of 616 patients were randomly divided into two groups (410 receiving pembrolizumab plus pemetrexed-platinum, 206 receiving placebo plus pemetrexed-platinum); the median time from randomisation to the data cut-off date of March 8, 2022, was 646 months (ranging from 601 to 724 months).
